Bilaga A. Protokollsavbrott: Förvaring och omtestning av värmebehandlade lysater
1. Förvara lyseringsprovröret med ett rent lock (se "LYSERING", 4.5) för att förvara ett värmebehandlat lysat.
2. Förvara i 4 till 8°C i upp till 72 timmar.
3. Förbered ett prov som har förvarats för amplifiering genom att vända det upp och ned 2–3 gånger så att det blandas.
4. Ta bort locken från rören.
5. Placera de omblandade lysatprovrören i 3M Molekylär Detektion, Insats för Värmeblock och värm vid 100 ±1 °C i 5 ±1
minuter.
6. Ta bort de icke övertäckta ställen med LL-provrör från uppvärmningsblocket och låt det svalna i 3M Molekylär
Detektion, Insats för kylblock i åtminstone 5 minuter och maximalt 10 minuter.
7. Fortsätt protokollet från avsnittet "Amplifiering" som beskrivs ovan.
