Use of Normal-Phase and HILIC columns in
Supercritical Fluid Chromatography (SFC)
Agilent columns suitable for SFC are typically normal-phase or HILIC columns
and are shipped in either normal-phase or HILIC mobile phases. Poroshell
120 EC-CN columns are shipped in reversed-phase solvent. Column pressure
and pH limits remain valid in SFC. Please keep in mind that pH in SFC is not
directly comparable with pH values of aqueous solutions. Therefore, the
apparent pH in CO
/organic modifier might differ.

Column Equilibration
Prior to use, flush column with 10-30 volumes of Methanol or isopropyl
alcohol to completely flush out shipping solvents. Then, equilibrate column
with approximately 10-20 column volumes of mobile phase at starting
conditions.
Column Storage
Make sure that you completely remove additives such as acids, bases
or buffers to prevent damage to the column. Flush the column with
10–20 column volumes of methanol or isopropyl alcohol to remove all
carbon dioxide. Do not store columns in a mixture of CO
and MeOH as CO
2

will evaporate and dry out the stationary phase. Properly seal with end plugs
to prevent solvent evaporation.
