6 System Operation
6.1 Preparing for a Run
6.1.1 Cryowedge
To prepare the Cryowedge for use:
Remove the upper half of the Cryowedge insulated cover.

Remove the wing nuts holding the transparent cover and set aside.
t
Remove the transparent cover. Fill the Cryowedge with the test solution
t
using a graduated cylinder. Standard volumes for each size Cryowedge
are as follows:

CW125 (20 inch)
CW300 (30 inch)
CW300 (34 inch)
Replace the transparent cover.

Replace the wing nuts and tighten them hand tight. Do not over-torque
t
or strip the threads.
Replace the upper half of the Cryowedge insulated cover.
t
Ensure that the Cryomixer Jr. cable is plugged into the Cryomixer Jr. and
t
the computer.
Ensure that at least six thermo-couples are plugged into the data
t
acquisition device, starting with position 1.
Place five of the thermocouples through the pre-drilled holes in the
t
transparent cover. Secure the thermocouples ?" below the surface of the
test solution using plastic spacers or cut pieces of silicon tubing.
Place the remaining thermocouple on the top of the transparent cover
t
as a reference temperature at the outer surface of the Cryowedge.
Record the number and position of each thermocouple for later reference.

6.1.2 S³ Module
Celsius Paks are delivered pre sterilized by gamma irradiation. They are sup-
®
plied with female Luer fittings and caps on 2 fill|drain ports. A variant with C-
Flex tubing extensions is also available for sterile filling. All Celsius
®
have a closed silicone/EVA thermowell for insertion of a thermocouple
probe.
In order to obtain consistent results, always load the S³ Module with the
same number of Celsius
®
same volume. The thermocouple should be placed at the same depth every
time.
