Washing process

First washing cycle

A washing process consists of several successive automatic current wash cycles. The number of wash cycles is
variable.
A washing cycle consists of 3 to 5 steps:
1. FILL
Filling in the physiological saline solution.
The tubes are filled with physiological saline solution at a rotation speed of 1100 RPM. The number of
revolutions per minute is not adjustable. The physiological saline solution is injected directly into the tubes, in
order to receive a good resuspension of the cells.
2. SPIN
Sedimentation.
The erythrocytes are sedimented by a selectable rotation speed. The time only begins to count after the set
rotation speed is reached. After the time has elapsed, a quick braking follows to prevent a resuspension of the
pellet.
3. DCNT
Decanting.
The excess is decanted at a selectable rotation speed. During decanting, the rotor turns in the opposite direction
of rotation.
4. DOWN
Centrifugation run to centrifuge the remaining droplets on the side of the tube to the bottom of the tube. The
speed is 2000 RPM and can not be adjusted.
This centrifugation run can be selected and deselected.
Selecting centrifugation run:
In the global parameters D.SpinDown (s) adjust the running time and set the program parameter DSpinDown to
"ON".
Deselect centrifugation run:
Either set the global parameter D.SpinDown (s) to "0" or in the global parameter D.SpinDown (s) adjust the
running time and set the program parameter DSpinDown to "OFF".
5. AGIT
Shaking.
The pellet is loosened again for the subsequent wash cycle by a fast, brief movement of the rotor.
In order not to loosen the pellet, no shaking occurs during a washing process with only one wash cycle or during
the last wash cycle of a washing process.
Select shaking:
Either in the global parameters Shakings select the number of shaking movements, or in the global parameters
ShakeT (min) adjust the duration of the shaking.
Deselect shaking:
Set the global parameter Shakings to "0".
