Comparaison d'intensité - Intensity comparison
Si vous souhaitez faire une étude d'intensité sur vos images, faites d'abord vos réglages (laser,
gain) sur l'échantillon le plus brillant et gardez ces paramètres pour acquérir les images
suivantes. Ainsi, les autres échantillons ne seront pas saturés. De même, les améliorations
d'images (contraste par exemple) doivent d'abord être faites sur l'image la plus brillante puis
appliquer de la même manière sur les autres images.
If you want to do an intensity analysis on your images, first adjust your settings (laser, gain)
on the brightest sample and keep these settings to acquire the following images. This way,
the other samples will not be saturated. In the same way, image enhancements (averaging
for example) should be made on the brightest image first and then applied in the same
method to the other images.

Dans Acquisition Mode, vérifier qu'on est en XYZ -
we are in XYZ

Allez dans l'onglet Z-stack qui est en bas à gauche -

Cliquer sur Live -

Déplacez-vous en z à l'aide du joystick ou du bouton du panel pour déterminer le 1er plan

Cliquez sur "Begin" dans Z-stack -

Déplacez-vous en z à l'aide du joystick ou du bouton du panel pour déterminer le dernier
plan -
Move in z with the joystick or the panel button to determine the last plane

Cliquez sur "End" dans Z-stack -

Déterminez le nombre de plans à acquérir ou l'intervalle entre les plans. pour avoir un
échantillonnage selon Nyquist, sélectionnez "System optimized" -
plans to be acquired or the interval between plans. To have Nyquist sampling, select
"System optimized".
Click on Live
Click on 'Begin' button in Z-stack tab
Click on 'End' button in Z-stack tab
