5.2 Wrap the second closing cap in aluminum foil, and
autoclave the assembled filter holder as well as the wrapped
closing cap for 30 minutes at 121°C.
5.3 Perform sterilizing filtration as you would pressure
filtration. Observe the usual precautions to avoid
contamination.
5.4 After completing filtration, shut off pressure source,
and remove the nipple along with the attached pressure
hose; simply pull off nipple from the connection (Fig. 13).
5.5 Unscrew the entire upper portion of the filter holder
and remove (Fig 14).
5.6 Cap the receiver flask with the other autoclaved closing
cap (Fig. 15).

6. Microbiological Testing

6.1 Insert the appropriate membrane filter, 47 mm d,
into the filter holder.
6.2 Plug all connections on the top and bottom lids
using the plugs provided.
6.3 Insert a plug into one of the lateral connections,
and cover the other with aluminum foil.
6.4 Screw filling funnel onto the filling port and cover
with aluminum foil.
6.5 Now the filter holder is ready to be autoclaved
at 121°C for 30 min. (Fig. 16).
6.6 After autoclaving, remove the aluminum foil. Connect
the filter holder to a vacuum source by attaching a hose
nipple to one of the lateral connections and connecting
a hose to the nipple and vacuum source.
6.7 Pour the liquid to be filtered into the filling funnel
and apply vacuum to the filter holder to begin filtration
(Fig. 17).
6.8 After completing filtration, pull the hose nipple along
with the hose off the connection, and remove the upper
portion of the filter holder (Fig. 18).
6.9 Remove the membrane filter from the filter rest using
sterile forceps. Incubate it on a nutrient pad, on agar, or
in other culture media (Fig. 19).

7. Sterility Testing

7.1 You may attach the upper portion only of SM 16511
to a vacuum manifold (SM 16826, 3-place), and perform
the sterility test (Fig. 20).
